preparation | sodium 60% hydride (5g, 0.125 mol) was added to the stirred Cbz-L-valine (9.5g, 0.038 mol) in the mixture of anhydrous THF (150 mL) and stir for 30 minutes at the same time. Add methyl iodide (20mL,0.32mol) and stir the obtained mixture at room temperature overnight. Ice water (200mL) was poured into the mixture at 0°C, and the resulting mixture was extracted with ether (2x 200mL). The aqueous layer was acidified to pH 1.5 using dilute HCl solution and extracted with ethyl acetate (3x 200mL). The combined organic extract was washed with saturated sodium thiosulfate solution (100 mL), dried (Na2SO4) and evaporated in vacuum to obtain N-benzyloxycarbonyl-N-methyl-L-valine (5g,55%), which is a colorless viscous oil and can be used in the next step without purification. |
